BODIPY?493/503 methyl bromide is a BODIPY dye. BODIPY dye is a small molecule dye with strong ultraviolet absorption ability, its fluorescence peak is relatively sharp, and the quantum yield is high. They are relatively insensitive to the polarity and pH of the environment and are relatively stable under different physiological conditions. Due to its structural asymmetry, BODIPY derives a variety of structural products. BODIPY lipid droplet dyes can well pass through the cell membrane into the cell, and localize the polar lipids in the cell to specifically stain the lipid droplets, which can be used for labeling of live cells and fixed cells. Maximum excitation/emission wavelength: 493/503 nm. Protect from light, stored at -20℃ for 1 year.

使用方法
1. BODIPY?493/503 methyl bromide 工作液的配制
1.1 制备储存液
用 382 μL 无水 DMSO 稀释 1mg BODIPY?493/503 methyl bromide 去配制 10 mM 的储备液。
注：BODIPY?493/503 methyl bromide 储存液建议分装后于 -20℃ 或 -80℃ 避光保存。
1.2 工作液的配制
用预热好的无血清细胞培养基或 PBS 稀释储存液，配制成 1-10 μM 的 BODIPY?493/503 methyl bromide 工作液。
注：请根据实际情况调整 BODIPY?493/503 methyl bromide 工作液浓度，且现用现配。
2. 细胞染色（悬浮细胞）
2.1 离心收集细胞，加入 PBS 洗涤两次，每次 5 min。细胞密度在 1×10⁶/mL
2.2 加入 1 mL BODIPY?493/503 methyl bromide 工作液，室温孵育 5-30 min。
2.3 400 g，离心 3-4 min，弃去上清。
2.4 加入 PBS 洗涤细胞两次，每次 5 min。
2.5 用 1 mL 无血清培养基或 PBS 重悬细胞后，使用荧光显微镜或流式细胞仪进行观察。
3. 细胞染色（贴壁细胞）
3.1 将贴壁细胞培养于无菌盖玻片上。
3.2 从培养基中移出盖玻片，吸除多余培养基。
3.3 加入 100 μL 染料工作液，轻轻晃动使其完全覆盖细胞，孵育 5-30 min。
3.4 吸去染料工作液，用培养基洗 2-3 次，每次 5 min，使用荧光显微镜或流式细胞仪进行观察。
4.切片染色
4.1 石蜡切片
4.1.1 室温脱蜡、水化：Xylene I 10 min、Xylene II 8 min、Xylene III 8 min、无水乙醇 5 min、90% 乙醇 3 min、80% 乙醇 3 min、70% 乙醇 3 min。蒸馏水 3 min*3 次，PBS 3 min*3 次
4.1.2 热抗原修复，换新的切片架，放入水化后的切片。加入抗原修复液，加热煮沸。温度控制在 96-98℃。然后将切片放入热锅中 15 min，最后自然冷却室温。PBS 5 min*2 次，蒸馏水 3 min*2 次。
4.1.3 免疫组化笔画圈：取出组织，甩干水分，可用吸水纸吸干水珠。用组化笔画圈围住组织，圆圈完整。
4.1.4 灭活内源酶活性：将切片放入湿盒中，盒中加入少量蒸馏水，滴加 3% 过氧化氢，室温孵育 10 min。PBS 3 min*3 次，蒸馏水水洗 3 min。
4.1.5 在组织中加入 50-100 μM 染料，孵育 30-60 min。
4.1.6 吸去染料工作液，用 PBS 或培养基洗 2-3 次，每次 5 min。
4.1.7 使用荧光显微镜观察进行成像观察。

保存条件
-20℃ 避光保存一年。

注意事项
1. 请根据实际情况调整 BODIPY?493/503 methyl bromide 工作液浓度与孵育时间。
2. 实验建议进行阳性对照，将对照组细胞与 30 μM 油酸孵育 8 h 后进行后续实验。
