HDACs/mTOR Inhibitor 1 是一种双重的 HDACs 和 mTOR 抑制剂，对 HDAC1, HDAC6, mTOR 的 IC₅₀s 分别为 0.19 nM、1.8 nM、1.2 nM。HDACs/mTOR Inhibitor 1 刺激细胞周期停滞在 G0/G1 期并诱导肿瘤细胞凋亡，且 in vivo 毒性低。HDACs/mTOR Inhibitor 1 可用于血液系统恶性肿瘤的研究。

生物活性

HDACs/mTOR Inhibitor 1 is a dual HDACs and mTOR inhibitor, with IC₅₀s of 0.19 nM, 1.8 nM, 1.2 nM for HDAC1, HDAC6, mTOR, respectively. HDACs/mTOR Inhibitor 1 stimulates cell cycle arrest in G0/G1 phase and induces tumor cell apoptosis with low toxicity in vivo. HDACs/mTOR Inhibitor 1 can be used in the research of hematologic malignancies.

体外研究

HDACs/mTOR Inhibitor 1 (Compound 12l) 抑制白血病细胞增殖，对 MV4-11, OCI-AML2, OCI-AML3IC 的₅₀s 分别为 4.05, 9.01, 9.98 μM。
HDACs/mTOR Inhibitor 1 (0-10 μM, 6 h or 24 h) 在 MV4-11 细胞中下调 p-ERK 水平，上调 Ac-H3 水平。
HDACs/mTOR Inhibitor 1 (10 to 1000 nM, 48 h) 减少 S 期 MV4-11 细胞细胞并增加 G0/G1 期细胞的百分比。

请依序添加每种溶剂： 10% DMSO 40% PEG300 5% Tween-80 45% saline
Solubility: ≥ 2.17 mg/mL (3.83 mM); Clear solution

此方案可获得 ≥ 2.17 mg/mL (3.83 mM，饱和度未知) 的澄清溶液。
以 1 mL 工作液为例，取 100 μL 21.7 mg/mL 的澄清 DMSO 储备液加到 400 μL PEG300 中，混合均匀；向上述体系中加入50 μL Tween-80，混合均匀；然后继续加入 450 μL生理盐水定容至 1 mL。
将 0.9 g 氯化钠，完全溶解于 100 mL ddH₂O 中，得到澄清透明的生理盐水溶液

此方案可获得 ≥ 2.17 mg/mL (3.83 mM，饱和度未知) 的澄清溶液，此方案不适用于实验周期在半个月以上的实验。
以 1 mL 工作液为例，取 100 μL 21.7 mg/mL 的澄清 DMSO 储备液加到 900 μL玉米油中，混合均匀。
