生物活性

Meclonazepam (Ro 11-3128) is a benzodiazepine compound with anxiolytic effect. Meclonazepam is active against immature S. mansoni. Meclonazepam (30 μM) eliminates 100% of immature parasites. Meclonazepam has a anti-schistosomal effect.

体外研究

Meclonazepam causes initially a massive spastic paralysis of immature S. mansoni.

Meclonazepam (0.1-5.6 mg/kg) exhibits anxiolytic effect in gerbils.
Meclonazepam (40 and 80 mg/kg) Has highly significant effect on the immature schistosomes.

Animal Model:	Adult experimentally naive male Mongolian gerbils (Meriones unguiculatus)
Dosage:	0.1, 0.3, 1.0, 3.0, 5.6 mg/kg
Administration:	Administration intraperitoneally (IP) 5 min prior to training onset.
Result:	The ED₅₀ of 0.77-0.78 mg/kg at the 5 min post-injection (p.i.).

Animal Model:	CB6F1 mice were infected with single-sex male S. mansoni cercariae 28 days before treatment
Dosage:	40 and 80 mg/kg
Administration:	Administered by gavage either as single drugs or in a sequence consisting of 1 dose of Praziquantel (PZQ), a 5-min interval, 1 dose of Ro, a 15-min interval, a second dose of PZQ.
Result:	PZQ had no significant effect on the immature schistosomes, whereas Ro 11-3128 was highly effective.
