D5D-IN-326 是一种选择性、可口服的 delta-5 desaturase (D5D) 抑制剂，在酶实验和细胞实验中，分别对大鼠和人 D5D 的 IC₅₀ 值为 72 和 22 nM，对 D6D 或 D9D 的活性无影响。在食物诱导的 C57BL/6J 肥胖小鼠中，D5D-IN-326 能够降低胰岛素耐受性，并且减轻小鼠体重。

D5D-IN-326 is a selective, orally active delta-5 desaturase (D5D) inhibitor, with IC₅₀s of 72 and 22 nM for rat and human D5D in enzymic and cell-based assays, respectively, has no effect on D6D or D9D activity. D5D-IN-326 reduces insulin resistance and decreases body weight in diet-induced obese C57BL/6J mice.

体内研究

D5D-IN-326 (0.1, 1, and 10 mg/kg, p.o. for 6 weeks) gradually decreases body weight at 10 mg/kg, but lowers doses are not effective in DIO mice after treatment for 6 weeks.
D5D-IN-326 (10 mg/kg, p.o. for 6 weeks) also significantly decreases gene expression levels of Ccl2, Cd68, Adgre1, and Il6, decreases epididymal Lep and Adipoq mRNAs which are significantly increased in HFD DIO mice.

请依序添加每种溶剂： 10% DMSO 40% PEG300 5% Tween-80 45% saline
Solubility: ≥ 2.08 mg/mL (4.39 mM); Clear solution

此方案可获得 ≥ 2.08 mg/mL (4.39 mM，饱和度未知) 的澄清溶液。
以 1 mL 工作液为例，取 100 μL 20.8 mg/mL 的澄清 DMSO 储备液加到 400 μL PEG300 中，混合均匀；向上述体系中加入50 μL Tween-80，混合均匀；然后继续加入 450 μL生理盐水定容至 1 mL。
将 0.9 g 氯化钠，完全溶解于 100 mL ddH₂O 中，得到澄清透明的生理盐水溶液

以 1 mL 工作液为例，取 100 μL 20.8 mg/mL 的澄清 DMSO 储备液加到 900 μL玉米油中，混合均匀。
