GSK180 是一种有效的、选择性和竞争性的犬尿氨酸-3-单加氧酶 (KMO) 抑制剂，它是色氨酸代谢中的关键酶，IC₅₀ 值约为 6 nM。GSK180 对色氨酸途径上其他酶的作用可忽略。GSK180 可快速改变犬尿氨酸 (kynurenine) 通路代谢物的水平，是动物模型中探索 KMO 抑制剂的有效工具。

生物活性

GSK180 is a selective, competitive, and potent inhibitor of kynurenine-3-monooxygenase (KMO), a key enzyme of tryptophan metabolism (IC₅₀, ~6 nM), but shows negligible activity against other enzymes on the tryptophan pathway. GSK180 rapidly changes levels of kynurenine pathway metabolites, and acts as a useful tool to probe the therapeutic potential of KMO inhibition.

体外研究

GSK180 inhibits endogenous KMO activity in primary human hepatocytes (IC₅₀=2.6 µM). GSK180 inhibits rat KMO slightly less potently than the human enzyme (IC₅₀=7 µM).

请依序添加每种溶剂： 10% DMSO 40% PEG300 5% Tween-80 45% saline
Solubility: ≥ 2.08 mg/mL (7.53 mM); Clear solution

此方案可获得 ≥ 2.08 mg/mL (7.53 mM，饱和度未知) 的澄清溶液。
以 1 mL 工作液为例，取 100 μL 20.8 mg/mL 的澄清 DMSO 储备液加到 400 μL PEG300 中，混合均匀；向上述体系中加入50 μL Tween-80，混合均匀；然后继续加入 450 μL生理盐水定容至 1 mL。
将 0.9 g 氯化钠，完全溶解于 100 mL ddH₂O 中，得到澄清透明的生理盐水溶液

将 2 g 磺丁基醚 β-环糊精加入 5 mL 生理盐水中，再用生理盐水定容至 10 mL，完全溶解，澄清透明
