IAXO-102

产品介绍

IAXO-102 是 TLR4 的拮抗剂，抑制 TLR4 信号通路。IAXO-102 抑制 MAPK 和 p65 NF-kB 的磷酸化及 TLR4 和 TLR4 依赖性的炎性蛋白的表达。IAXO-102 抑制实验性腹主动脉瘤的发展。

生物活性

IAXO-102 is a TLR4 antagonist which negatively regulates TLR4 signalling. IAXO-102 inhibits MAPK and p65 NF-κB phosphorylation and expression of TLR4 dependent proinflammatory protein. IAXO-102 also prevents experimental abdominal aortic aneurysm development.

体外研究

IAXO-102 (1-10 µM, for 2 hours) inhibits MAPK and p65 NF-κB phosphorylation in human umbilical vein endothelial (HUVEC) cells.
IAXO-102 (10 µM, for 17 hours) suppresses LPS induced proinflammatory proteins MCP-1 and IL-8 production in HUVEC.

Western Blot Analysis

Cell Line:	Human umbilical vein endothelial (HUVEC) cells
Concentration:	1-10 µM
Incubation Time:	Pretreatment for 1 hour and then exposed to LPS (100 ng/mL) for additional 1 hour
Result:	Significantly inhibited LPS-stimulated MAPK/p65nF-ΚB phosphorylation.

请依序添加每种溶剂： 10% EtOH 40% PEG300 5% Tween-80 45% saline
Solubility: ≥ 0.83 mg/mL (1.42 mM); Clear solution

此方案可获得 ≥ 0.83 mg/mL (1.42 mM，饱和度未知) 的澄清溶液。
以 1 mL 工作液为例，取 100 μL 8.3 mg/mL 的澄清 EtOH 储备液加到 400 μL PEG300 中，混合均匀；向上述体系中加入50 μL Tween-80，混合均匀；然后继续加入 450 μL生理盐水定容至 1 mL。
将 0.9 g 氯化钠，完全溶解于 100 mL ddH₂O 中，得到澄清透明的生理盐水溶液

请依序添加每种溶剂： 10% EtOH 90% corn oil
Solubility: ≥ 0.83 mg/mL (1.42 mM); Clear solution

此方案可获得 ≥ 0.83 mg/mL (1.42 mM，饱和度未知) 的澄清溶液，此方案不适用于实验周期在半个月以上的实验。
以 1 mL 工作液为例，取 100 μL 8.3 mg/mL 的澄清 EtOH 储备液加到 900 μL玉米油中，混合均匀。
