产品介绍

Sepin-1 是一种有效的分离酶 (separase) 抑制剂，IC₅₀ 值为 14.8 µM。Sepin-1 抑制细胞增殖、迁移和伤口愈合。Sepin-1 降低 FoxM1 蛋白和 mRNA 水平的表达。Sepin-1 具有抗肿瘤活性。

生物活性

Sepin-1 is a potent separase inhibitor with an IC₅₀ value of 14.8 µM. Sepin-1 inhibits cell proliferation, migration and wound healing. Sepin-1 decreases the expression of FoxM1 protein and mRNA level. Sepin-1 shows anti-tumor activity.

体外研究

Sepin-1 (0, 20, 40 µM; 24 h) 抑制细胞迁移和伤口愈合，并引起 DNA 损伤。
Sepin-1 (0, 10, 20, 40 µM; 24 h) 以剂量依赖性方式降低 FoxM1 蛋白和 mRNA 水平的表达。

Cell Proliferation Assay

Cell Line:	BT-474, MCF7, MDA-MB-231, MDA-MB-468 cells
Concentration:	0-64 µM
Incubation Time:	3 days
Result:	Inhibited cell growth with EC₅₀s of 18.03, 17.66, 27.33, 27.92 µM for BT-474, MCF7, MDA-MB-231, MDA-MB-468 cells, respectively.

Western Blot Analysis

Solubility: ≥ 2.5 mg/mL (11.20 mM); Clear solution

此方案可获得 ≥ 2.5 mg/mL (11.20 mM，饱和度未知) 的澄清溶液。
以 1 mL 工作液为例，取 100 μL 25.0 mg/mL 的澄清 DMSO 储备液加到 900 μL 20% 的 SBE-β-CD 生理盐水水溶液中，混合均匀。
将 2 g 磺丁基醚 β-环糊精加入 5 mL 生理盐水中，再用生理盐水定容至 10 mL，完全溶解，澄清透明
